GI-4000 + gemcitabine is a combination therapy for Ras-mutant resected pancreatic cancer, consisting of GI-4000—a series of recombinant, heat-inactivated Saccharomyces cerevisiae yeast (tarmogen) vaccines expressing common mutated Ras proteins—and gemcitabine, a nucleoside analog chemotherapeutic. GI-4000 is designed to stimulate immune responses against mutated Ras proteins, aiming to enhance the cytotoxic activity against residual tumor cells after surgical resection. Each GI-4000 product is matched to the patient’s specific tumor Ras mutation. Gemcitabine works primarily through inhibition of DNA synthesis, inducing apoptosis in rapidly dividing cells. The combination leverages immunotherapeutic targeting of tumor-specific mutations and standard chemotherapy[1][4][5].
