GIP(1-16) is a C-terminally truncated peptide fragment of the 42-amino acid hormone glucose-dependent insulinotropic polypeptide (GIP). It consists of the N-terminal residues 1 through 16 of the native peptide. Research into GIP(1-16) was conducted to explore the development of small molecular weight GIP receptor ligands for metabolic disorders. While native GIP is a potent stimulator of insulin secretion, GIP(1-16) exhibits only weak agonist activity at the GIP receptor in vitro. In vivo studies in obese diabetic (ob/ob) mice demonstrated that GIP(1-16) has minimal impact on insulin release and does not significantly improve glucose tolerance, leading to the conclusion that the C-terminal region of GIP is essential for biological activity and that GIP(1-16) lacks therapeutic potential.
