Overview

GIP(3-30), also known as GIP(3-30)NH2, is a naturally occurring, truncated peptide fragment of glucose-dependent insulinotropic polypeptide (GIP) that acts as a potent and selective competitive antagonist of the GIP receptor (GIPR). Discovered and characterized by researchers at the University of Copenhagen and Gentofte Hospital, GIP(3-30) represents the first GIPR antagonist administered to humans. It has been widely utilized as a clinical research tool and physiological probe to elucidate the role of endogenous GIP in glucose homeostasis, insulin secretion, bone resorption, splanchnic blood flow, and conditions such as postprandial hypotension, postural orthostatic tachycardia syndrome (POTS), and acromegaly. While it serves as a foundational molecule for GIPR antagonist drug design, commercial therapeutic development has shifted toward optimized analogs like AT-7687.
