Overview

Giredestrant + ipatasertib is an investigational combination therapy being studied for the treatment of breast cancer. Giredestrant is a next-generation oral selective estrogen receptor degrader (SERD) that binds to and induces degradation of the estrogen receptor (ER), thereby inhibiting ER signaling in ER-positive breast cancer cells. Ipatasertib is an oral small molecule inhibitor of AKT (protein kinase B), a key component of the PI3K/AKT signaling pathway involved in cell proliferation and survival. The combination aims to target both hormone-driven and PI3K/AKT pathway-driven tumor growth mechanisms. This regimen is under investigation primarily for patients with inoperable, locally advanced or metastatic ER-positive breast cancer who have progressed after prior cyclin-dependent kinase 4/6 inhibitor therapy[1][2][7].
