**glembatumumab vedotin + CDX-301** is an experimental combination therapy investigated primarily for advanced melanoma. Glembatumumab vedotin is an antibody-drug conjugate targeting glycoprotein NMB (gpNMB), consisting of a fully human monoclonal antibody linked to monomethyl auristatin E (MMAE), a potent microtubule inhibitor. It binds gpNMB expressed on tumor cells, delivers MMAE intracellularly, and induces cell death through cytotoxicity and bystander effect. CDX-301 (also known as FLT3 ligand) is a recombinant human protein that stimulates the production and maturation of dendritic cells and other hematopoietic cells, enhancing immune responses against tumor cells. This combination aims to directly kill cancer cells and augment antitumor immunity by increasing dendritic cell numbers and function.
