A combination therapy of **glembatumumab vedotin**, an antibody-drug conjugate targeting glycoprotein NMB (gpNMB), and **pembrolizumab**, a humanized monoclonal antibody targeting the programmed cell death-1 (PD-1) receptor. Glembatumumab vedotin delivers the cytotoxic agent monomethyl auristatin E (MMAE) to gpNMB-expressing tumor cells to induce cell death. Pembrolizumab blocks PD-1, enabling T cells to mount an anti-tumor immune response. This combination was investigated for advanced melanoma, but development was discontinued after disappointing trial results.
