Overview
Glembatumumab vedotin + varlilumab is an investigational combination therapy for advanced melanoma and other cancers. Glembatumumab vedotin is an antibody-drug conjugate (ADC) that targets glycoprotein NMB (gpNMB), a transmembrane protein associated with poor prognosis in several cancers. It consists of a fully human IgG2 monoclonal antibody (CR011) linked to the cytotoxic agent monomethyl auristatin E (MMAE). Upon binding to gpNMB-expressing tumor cells and internalization, MMAE is released intracellularly to inhibit microtubule formation and induce cell death[1][2][6]. Varlilumab is a fully human monoclonal agonist antibody targeting CD27, a co-stimulatory molecule on T cells. By activating CD27, varlilumab enhances T cell activation and proliferation as part of the immune response against tumors[8]. The combination aims to leverage direct tumor cell killing by glembatumumab vedotin with immune system activation by varlilumab. Clinical trials have evaluated this combination in patients with advanced melanoma[4][7].
