Overview

Glimepiride + insulin glargine is a combination therapy used in the management of type 2 diabetes mellitus (T2DM), particularly for patients whose blood glucose levels are not adequately controlled with oral antidiabetic drugs alone. Glimepiride is a second-generation sulfonylurea that stimulates insulin secretion from pancreatic beta cells by blocking ATP-sensitive potassium channels, thereby increasing endogenous insulin release. Insulin glargine is a long-acting recombinant human insulin analog that provides basal insulin coverage for up to 24 hours or longer, helping to maintain stable blood glucose levels throughout the day and night. The combination aims to improve glycemic control by addressing both insufficient endogenous insulin secretion and providing exogenous basal insulin support. Clinical studies have shown this combination can significantly reduce HbA1c and fasting plasma glucose without increasing severe hypoglycemia risk when appropriately monitored[2][3][4][5][6].
