GLIX1 + PARPi is an experimental combination therapy being developed by Hemispherian for the treatment of glioblastoma (GBM). The combination consists of GLIX1, a small molecule activator of the TET2 (Ten-Eleven Translocation 2) enzyme, and a PARP (Poly ADP-ribose polymerase) inhibitor. GLIX1 works by restoring levels of 5-hydroxymethylcytosine (5hmC), an epigenetic mark that is frequently lost in cancer cells, which increases their susceptibility to DNA damage. By inhibiting PARP-mediated DNA repair in this sensitized state, the combination aims to enhance the accumulation of lethal DNA lesions specifically within tumor cells. This therapeutic strategy is designed to overcome resistance in high-grade gliomas and is currently being evaluated in Phase 1/2 clinical studies.
