Overview
GLPG4716 + midazolam is a combination of **GLPG4716** (also known as OATD-01), a small molecule inhibitor of chitinase enzymes, and **midazolam**, a short-acting benzodiazepine sedative. GLPG4716 targets **chitotriosidase** and **acidic mammalian chitinase** (CHIT1/AMCase), and is under development mainly for treatment of idiopathic pulmonary fibrosis and other fibrotic diseases. Midazolam is an established benzodiazepine acting as a positive allosteric modulator of the **GABA-A receptor**, used for sedation, anesthesia, anticonvulsant therapy, and muscle relaxation. The combination is being investigated primarily in clinical pharmacology studies for potential drug-drug interactions, particularly the impact of GLPG4716 on the pharmacokinetics of midazolam, with relevance to cytochrome P450 3A4 modulation[1][2][6][7].
