Overview

This is a **combination vaccine formulation** developed as a prophylactic for herpes simplex virus type 2 (HSV-2), comprising **truncated recombinant glycoprotein D (gD-2t) from HSV-2**, **aluminum hydroxide (alum)** as an adjuvant, and **3-O-deacylated monophosphoryl lipid A (MPL)** as an immune enhancer[2][5]. Glycoprotein D is a major viral envelope protein fundamental for HSV-2 entry into host cells and the dominant target for virus-neutralizing antibodies and T-cell responses[2][3]. MPL, a detoxified derivative of bacterial lipid A, activates innate immunity through Toll-like receptor 4, promoting a robust adaptive immune response when combined with alum. This vaccine induces high titers of binding and neutralizing antibodies, and enhances both cell-mediated (CD4+ and CD8+ T cell) and humoral immunity against HSV-2[2][3]. Developed primarily for the prevention of genital herpes, it has completed multiple clinical trials, including large efficacy studies. While it elicits a strong immune response, efficacy against HSV-2 infection has not been consistently observed, but there has been significant efficacy against HSV-1 infection and genital disease in certain subpopulations[2][5].
