A combination vaccine formulation consisting of **glycoprotein E** (gE) antigen, **CpG 1018** (a synthetic Toll-like receptor 9 [TLR9] agonist oligonucleotide adjuvant), and **alum** (aluminum hydroxide adjuvant). This combination is under development notably as a recombinant subunit vaccine against **herpes zoster (shingles)**, and studied in other vaccine candidates. **gE** serves as the immunogen; **CpG 1018** functions as an immunostimulatory adjuvant to promote a Th1-biased immune response; and **alum** further enhances immunogenicity. The combination has demonstrated dose-sparing effects and robust induction of antibody and cellular immunity[1][2][4][5]. CpG 1018 is also FDA-approved as an adjuvant in the hepatitis B vaccine HEPLISAV-B but is used here in combination with alum for potentially greater immunogenicity in other vaccine platforms[1][3][5].
