This is a combination immunotherapy regimen consisting of granulocyte-macrophage colony-stimulating factor (GM-CSF), interleukin-2 (IL-2), and the monoclonal antibody mAb17-1A. The monoclonal antibody mAb17-1A targets colorectal carcinoma cells, while GM-CSF and IL-2 are cytokines that modulate immune responses. The rationale for this combination is to enhance anti-tumor immunity by increasing antibody-dependent cellular cytotoxicity (ADCC) and stimulating immune effector cells. Clinical studies in patients with metastatic colorectal cancer have shown that adding IL-2 to the regimen of GM-CSF plus mAb17-1A increases certain immune activation markers but may also induce an immunosuppressive state compared to GM-CSF alone, affecting cytotoxic mononuclear cells and B cell function[1]. This approach has been investigated primarily in clinical trial settings.
