**Gocatamig + durvalumab** is an investigational combination therapy for relapsed or refractory extensive-stage small cell lung cancer (ES-SCLC). Gocatamig (also known as MK-6070 or HPN328) is a T-cell engager developed using the TriTAC platform that targets Delta-like ligand 3 (DLL3) on tumor cells and CD3 on T cells, promoting T cell-mediated tumor cell lysis. Durvalumab is a monoclonal antibody checkpoint inhibitor that targets programmed death ligand 1 (PD-L1), preventing T-cell exhaustion and enhancing anti-tumor immunity. The combination is being evaluated for synergistic effects due to complementary mechanisms and minimally overlapping toxicities, aiming to improve efficacy without significantly increasing adverse events. The combination is being tested in ongoing phase 1b/2 studies for the treatment of ES-SCLC[1][2][3].
