Overview

Golcadomide + roginolisib is an investigational, fully oral combination regimen comprising golcadomide, a cereblon E3 ligase modulator (CELMoD) that induces proteasomal degradation of transcription factors such as Ikaros and Aiolos, and roginolisib, a highly selective phosphatidylinositol 3-kinase delta (PI3Kδ) inhibitor that suppresses PI3K/AKT signaling and promotes apoptosis in malignant lymphoid cells.[4][8] Golcadomide is being developed by Bristol Myers Squibb as a next-generation immunomodulatory agent for non-Hodgkin lymphomas and T‑cell lymphomas, while roginolisib (IOA‑244) is being developed by iOnctura with ongoing trials in uveal/ocular melanoma and hematologic malignancies.[6][8] The golcadomide + roginolisib combination is under exploration in platform and combination studies for relapsed or refractory peripheral T‑cell lymphoma and other lymphomas, where it is often combined with additional agents such as azacitidine or venetoclax to achieve synergistic antitumor activity through concurrent modulation of cereblon-dependent transcriptional programs and PI3Kδ‑driven survival pathways.[4][14][16]
