Gold nanoparticles + Lys05 is an experimental combination therapy consisting of the potent lysosomotropic agent Lys05—a dimeric derivative of chloroquine—and gold nanoparticles (AuNPs) serving as a delivery vehicle. Lys05 acts as an autophagy inhibitor by accumulating within lysosomes, where it raises the intralysosomal pH and impairs the function of acid hydrolases, thereby blocking the fusion of autophagosomes with lysosomes and preventing the degradation of cellular components. Specifically, Lys05 has been identified as an inhibitor of Palmitoyl-protein thioesterase 1 (PPT1), a key enzyme in lysosomal protein degradation. The integration of Lys05 with gold nanoparticles is designed to enhance the cellular uptake and lysosomal targeting of the inhibitor, potentially overcoming resistance mechanisms in various malignancies such as melanoma and glioblastoma. This approach leverages the unique physicochemical properties of AuNPs to improve the therapeutic index of autophagy inhibition in oncology.
