golimumab + methotrexate

Golimumab + methotrexate is a combination therapy used primarily for the treatment of moderately to severely active rheumatoid arthritis (RA) in adults and children, as well as other inflammatory arthritides. Golimumab is a fully human monoclonal antibody that targets and neutralizes tumor necrosis factor alpha (TNF-α), a key cytokine involved in systemic inflammation. Methotrexate is a conventional synthetic disease-modifying antirheumatic drug (DMARD) that acts mainly by inhibiting dihydrofolate reductase, interfering with folate metabolism, and exerting anti-inflammatory effects through multiple mechanisms including increased adenosine signaling. The combination of these two agents has demonstrated superior efficacy compared to either agent alone in reducing signs/symptoms of RA, limiting radiographic progression of joint damage, and improving physical function[1][2][3][4][5][7][9]. This regimen is recommended for patients who have had an inadequate response to methotrexate monotherapy or other DMARDs.
