Overview

A combination therapy comprising goserelin, a synthetic decapeptide and gonadotropin-releasing hormone (GnRH) agonist, and docetaxel, a taxane-derived antineoplastic agent. This regimen is being investigated by Peking Union Medical College in a Phase 2 umbrella trial for patients with androgen receptor (AR)-positive salivary gland carcinoma. Goserelin functions by initially stimulating and subsequently desensitizing GnRH receptors in the pituitary gland, leading to a profound reduction in serum testosterone levels. Docetaxel complements this by binding to and stabilizing tubulin, which inhibits microtubule reorganization and arrests the cell cycle in the G2/M phase, ultimately inducing apoptosis. The combination aims to leverage both hormonal deprivation and cytotoxic effects to treat advanced salivary malignancies.
