gp100(25-33) is a 9-amino acid peptide fragment derived from the glycoprotein 100 (gp100) protein, a lineage-specific antigen expressed in melanocytes and highly overexpressed in melanoma. This specific sequence (EGSRNQDWL in mice; KVPRNQDWL in humans) serves as a cytotoxic T lymphocyte (CTL) epitope restricted by MHC class I molecules. In cancer immunotherapy research, the peptide is used as a vaccine component to prime and expand antigen-specific CD8+ T cells. It is frequently studied in preclinical melanoma models, where it may be administered with adjuvants like CpG, pulsed onto dendritic cells, or conjugated to carrier cells using crosslinkers such as sulfo-SMCC to enhance immunogenicity and targeted delivery to lymphoid tissues.
