The gp100:154-162 Peptide (KTWGQYWQV) is a naturally processed epitope derived from the melanocyte-specific gp100 protein. This peptide is recognized by HLA-A*0201-restricted, melanoma-reactive cytotoxic T lymphocytes (CTLs)[1][2]. It appears to be the most abundant peptide at the cell surface among several gp100-derived epitopes[1][7]. ## Description The gp100:154-162 Peptide is a nonamer (9-amino acid peptide) with the sequence KTWGQYWQV that corresponds to amino acids 154-162 of the gp100 protein[1][2]. This peptide is presented by HLA-A*0201 molecules on the surface of melanoma cells and normal melanocytes, making it a target for immunotherapy approaches against melanoma[1][2][3]. It has been identified as a dominant epitope recognized by tumor-infiltrating lymphocytes (TILs) from melanoma patients, particularly TIL 1200[2]. Studies have shown that a 100-fold lower concentration of this peptide was required for target-cell sensitization compared to other gp100 peptides, indicating its potency as an immunogenic epitope[2]. ## Development and Clinical Applications The gp100:154-162 Peptide has been developed by the National Cancer Institute (NCI) as a potential therapeutic vaccine for melanoma[6]. It has been used in clinical trials, particularly for metastatic melanoma, often in combination with other treatments such as: - Adoptive T-cell therapy using genetically engineered lymphocytes - Peptide vaccines - Chemotherapy and radiation therapy Clinical trials have evaluated its efficacy in stimulating immune responses against melanoma cells, with some studies showing promising results in terms of T-cell activation and tumor recognition[3][6][8]. ## Mechanism of Action The peptide works by stimulating melanoma-specific cytotoxic T lymphocytes that can recognize and kill melanoma cells expressing the gp100 protein[1][2][7]. When presented by HLA-A*0201 molecules on antigen-presenting cells, the peptide activates CD8+ T cells that can then target melanoma cells displaying the same peptide-HLA complex on their surface. Studies in HLA-A*0201/Kb-transgenic mice have demonstrated that immunization with this peptide can elicit potent CTL responses capable of eliminating human melanoma cells both in vitro and in vivo[3][7].
