gp100 peptide + tyrosinase peptide is an investigational combination cancer vaccine therapy primarily developed for the treatment of malignant melanoma. It consists of synthetic peptides representing immunodominant epitopes derived from two melanoma-associated antigens: glycoprotein 100 (gp100, also known as PMEL) and tyrosinase. These peptides are typically modified to enhance their binding affinity to the HLA-A*0201 molecule, such as the g209-210M and tyr368-370D variants. The mechanism of action involves the presentation of these antigens by antigen-presenting cells to the immune system, thereby stimulating the expansion of antigen-specific CD8+ cytotoxic T lymphocytes (CTLs). These CTLs are then primed to recognize and destroy melanoma cells that express gp100 and tyrosinase on their surface. This combination is frequently administered with adjuvants such as Montanide ISA-51 and has been studied in conjunction with systemic cytokines like interleukin-2 (IL-2) to enhance the anti-tumor immune response.
