GPC1-ADC(MMAE) is an antibody-drug conjugate (ADC) consisting of a monoclonal antibody targeting glypican-1 (GPC1) conjugated to the microtubule-disrupting agent monomethyl auristatin E (MMAE). GPC1 is a cell-surface proteoglycan overexpressed in pancreatic ductal adenocarcinoma (PDAC) on both tumor cells and cancer-associated fibroblasts (CAFs). The ADC is designed to dual-target the tumor and its stroma. Upon binding to GPC1, the ADC is internalized; in CAFs, the released MMAE can be exported via MDR-1 to induce apoptosis in neighboring cancer cells, a mechanism that helps overcome the dense stroma characteristic of PDAC. It has shown robust preclinical efficacy in patient-derived xenograft (PDX) models.
