Overview

GPC2 + B7-H3 bicistronic CAR T cells is an experimental cell-based immunotherapy designed to treat pediatric solid tumors, specifically neuroblastoma and medulloblastoma. This therapy utilizes a bicistronic "OR" chimeric antigen receptor (CAR) construct that simultaneously targets glypican 2 (GPC2) and CD276 (B7-H3). Preclinical studies have demonstrated that these dual-targeting CAR T cells exhibit potent anti-tumor activity, enhanced persistence, and a less exhausted phenotype compared to single-target CARs. The development is led by researchers at institutions such as the Children's Hospital of Philadelphia, focusing on overcoming the interconvertible adrenergic and mesenchymal states in neuroblastoma where antigen expression can fluctuate.
