Overview

GPC3 CAR-T + TGFβ CAR-T is a combination cell therapy consisting of two distinct chimeric antigen receptor (CAR) T cell products. One component targets glypican-3 (GPC3), a cell surface proteoglycan highly expressed in hepatocellular carcinoma (HCC) and other solid tumors, while the other targets transforming growth factor beta (TGFβ), an immunosuppressive cytokine prevalent in the tumor microenvironment. The GPC3-targeted CAR-T cells are designed to recognize and kill tumor cells expressing GPC3, providing direct antitumor activity[1][5][9]. The TGFβ-targeted CAR-T cells are engineered to resist or convert the immunosuppressive effects of TGFβ into stimulatory signals for T-cell activation, thereby enhancing overall immune response against tumors and protecting effector cells from suppression[6][10]. This dual approach aims to overcome both antigen-driven tumor growth and microenvironment-mediated immune evasion, potentially improving efficacy in solid tumors such as HCC[7].
