Overview

GPC3-targeting CAR-T cells (Jianming Xu) is an investigational adoptive cellular immunotherapy consisting of autologous T cells genetically engineered to express a chimeric antigen receptor (CAR) specific for glypican-3 (GPC3). GPC3 is a cell-surface heparan sulfate proteoglycan that is highly overexpressed in hepatocellular carcinoma (HCC) but has minimal expression in healthy adult tissues, making it an ideal target for directed therapy. Upon administration, the CAR-T cells recognize GPC3-positive tumor cells, leading to T-cell activation, expansion, and targeted cytotoxicity against the cancer cells. This specific therapy was evaluated in a Phase 1 clinical trial (NCT05352542) sponsored by Jianming Xu for the treatment of patients with advanced, relapsed, or refractory hepatocellular carcinoma. The trial was designed to assess safety, tolerability, and efficacy but has since been terminated.
