Overview

GR1801 + rabies vaccine is a combination therapy under investigation for post-exposure prophylaxis (PEP) of rabies. The combination consists of GR1801, a novel human bispecific antibody, and a standard inactivated rabies vaccine. GR1801 is engineered by combining two monoclonal antibodies (A2 and B353) that target distinct epitopes on the rabies virus glycoprotein. It is an asymmetric immunoglobulin G1 molecule with one arm in fragment antigen-binding (Fab) form and the other as a single-chain variable fragment (scFv), constructed using Knobs-into-Holes technology. This design enables broad-spectrum neutralization against diverse natural variants of the rabies virus. In preclinical studies, GR1801 demonstrated protection equivalent to human rabies immune globulin (HRIG). The combination with the standard rabies vaccine follows WHO recommendations for PEP in high-risk exposures[1][2][4].
