Overview

GRm13Z40-2 + aldesleukin is a combination therapy consisting of two components: 1. **GRm13Z40-2**: An allogeneic, glucocorticoid receptor (GR)-negative cytotoxic T lymphocyte (CTL) product engineered to express a membrane-tethered interleukin 13 zetakine (IL13-Zetakine) and HyTK. These cells are designed to target the interleukin 13 receptor subunit alpha 2 (IL-13Rα2), which is overexpressed in certain brain tumors such as glioblastoma. The genetic modification also confers resistance to glucocorticoids, allowing these CTLs to remain active even in the presence of steroid treatment[1][4][5]. 2. **Aldesleukin**: A recombinant form of human interleukin 2 (IL-2), used as an immunostimulatory cytokine that enhances proliferation and activation of immune effector cells, including T cells and natural killer cells[6][8]. In this combination, aldesleukin is administered intracranially alongside GRm13Z40-2 to support the survival and function of infused CAR-T cells within the tumor microenvironment[5]. This combination has been investigated primarily for recurrent or refractory malignant gliomas, including glioblastoma multiforme and other high-grade brain tumors. Early-phase clinical trials have demonstrated safety and feasibility with some evidence of transient tumor reduction at infusion sites[4][5].
