Overview

This is an investigational **combination therapy** consisting of a peptide cancer vaccine (**GRN-1201**), granulocyte-macrophage colony-stimulating factor (**sargramostim**), and the human anti-PD-1 monoclonal antibody (**pembrolizumab**). - **GRN-1201** is a cancer peptide vaccine made of four HLA-A2-restricted peptides derived from four different tumor-associated antigens (TAAs), intended to induce a cytotoxic T-cell response against tumor cells expressing these antigens, notably in melanoma and non-small cell lung cancer. - **Sargramostim** (GM-CSF) acts as an immunostimulatory cytokine, enhancing the activation and proliferation of immune cells at the vaccination site. - **Pembrolizumab** is a monoclonal antibody that inhibits the programmed death receptor-1 (**PD-1**), blocking immune checkpoint signaling and thus enhancing T-cell mediated anti-tumor responses. The combination is designed to synergistically augment anti-tumor immunity, using the vaccine to present tumor antigens, GM-CSF to boost antigen presentation and immune cell recruitment, and pembrolizumab to release brakes on T-cell activity imposed by the tumor microenvironment. The combination has been studied primarily in **metastatic, PD-L1 positive non-small cell lung cancer** and resected melanoma[3][5][7][9].
