GRN163L (imetelstat) is a synthetic lipid-conjugated 13-mer oligonucleotide that acts as a telomerase inhibitor by targeting the RNA template region of human telomerase, thereby inhibiting its activity. Trastuzumab is a monoclonal antibody that targets the human epidermal growth factor receptor 2 (HER2), which is overexpressed in certain breast and gastric cancers. The combination of GRN163L and trastuzumab has been investigated for overcoming resistance to HER2-targeted therapy in HER2-positive breast cancer, with preclinical studies suggesting enhanced efficacy against cancer stem cell populations. GRN163L was developed by Geron, while trastuzumab was originally developed by Genentech/Roche and is available under several brand names[1][5][8].
