This combination therapy consists of recombinant human growth hormone (rhGH) and calcitriol, the active form of vitamin D3. It was investigated in a Phase II clinical trial sponsored by the National Center for Research Resources (NCRR) for the treatment of renal osteodystrophy in children and adolescents with end-stage renal disease (ESRD) on dialysis. Recombinant human growth hormone acts as an agonist at the growth hormone receptor, stimulating linear growth and bone formation, which is often impaired in pediatric renal failure. Calcitriol targets the vitamin D receptor to regulate calcium and phosphorus metabolism and suppress parathyroid hormone (PTH) secretion, addressing secondary hyperparathyroidism. The study aimed to evaluate whether the anabolic effects of growth hormone could improve bone health outcomes and counteract adynamic bone disease in this specific patient population.
