Overview

This entry refers to a combination therapy involving growth hormone and a gonadotropin-releasing hormone (GnRH) agonist. Growth hormone is a peptide hormone that stimulates growth, cell reproduction, and regeneration in humans. GnRH agonists are synthetic analogues of the natural hypothalamic neurohormone GnRH; they act as agonists at the gonadotropin-releasing hormone receptor on pituitary gonadotrophs, initially stimulating but then profoundly suppressing secretion of luteinizing hormone (LH) and follicle-stimulating hormone (FSH) through receptor desensitization[1][5][6]. This results in decreased production of sex steroids such as estrogen and testosterone. The combination is used clinically in certain pediatric endocrinology settings—for example, to optimize height outcomes in children with early puberty or specific growth disorders by delaying epiphyseal closure while promoting linear growth.
