This combination is an investigational personalized immunotherapy regimen composed of two patient-specific neoantigen vaccines (GRT-C901, an adenoviral vector; and GRT-R902, a self-amplifying mRNA vector, together branded as GRANITE) plus the immune checkpoint inhibitors nivolumab (a PD-1 inhibitor, IgG4 monoclonal antibody) and ipilimumab (a CTLA-4 inhibitor, IgG1 monoclonal antibody). The vaccination strategy uses a heterologous prime/boost approach (GRT-C901, then GRT-R902), designed to stimulate a robust T-cell response targeting patient-specific tumor neoantigens. Nivolumab and ipilimumab synergize with the vaccine by blocking co-inhibitory receptors to augment anti-tumor immunity. This regimen is under phase 2/3 clinical evaluation in metastatic colorectal cancer, particularly in the microsatellite-stable (MSS) subtype[1][3][5][7].
