Overview

**GS-4528 + zimberelimab** is an investigational combination therapy for advanced solid tumors. GS-4528 is a novel anti-TIGIT monoclonal antibody, while zimberelimab is an anti–programmed cell death protein 1 (PD-1) monoclonal antibody. The combination aims to enhance immunotherapy efficacy by targeting both the TIGIT and PD-1 pathways—two critical immune checkpoints involved in cancer immune evasion. Zimberelimab restores T-cell antitumor activity by blocking PD-1, and GS-4528 is believed to further augment T-cell responses by inhibiting the TIGIT pathway. This combination is being studied in early-phase clinical trials in patients with refractory or advanced solid tumors, with development led by Gilead Sciences and Arcus Biosciences[1][2][4][6][7][8][9].
