GS-4774 + OAV is a combination therapy for chronic hepatitis B comprising GS-4774, a heat-inactivated, yeast-based therapeutic vaccine (Tarmogen platform), and an oral antiviral (OAV; typically a nucleoside/nucleotide analogue antiviral such as tenofovir)[1][3][5][8]. The vaccine is designed to elicit hepatitis B virus-specific T cell responses by expressing HBV antigens from major genotypes, aiming to aid the immune system in eliminating infected cells, while the OAV component suppresses viral replication. The combination is intended to enhance seroconversion rates and potentially provide functional cure for chronic HBV infection[3][6]. \nGS-4774 was developed by GlobeImmune and licensed to Gilead Sciences[3][6]. Clinical trials have shown safety and tolerability, but as of the most recent studies, the combination did not achieve significant reductions in HBsAg or lead to clinical cure in virally suppressed patients; some benefit was seen in specific patient subgroups, and further studies in treatment-naïve populations are ongoing[1][6][8].
