Overview

**GS-9190 + GS-9256** is an investigational oral combination therapy for chronic hepatitis C virus (HCV) infection, particularly genotype 1. GS-9190 (also known as tegobuvir) is a non-nucleoside inhibitor of the HCV NS5B RNA-dependent RNA polymerase, while GS-9256 is a NS3 serine protease inhibitor. This combination, developed by Gilead Sciences, was studied both as a direct-acting oral antiviral regimen and in combination with ribavirin and pegylated interferon. Clinical studies indicated that adding ribavirin or ribavirin/pegylated interferon to the combination enhanced antiviral activity, with significant declines in HCV RNA and substantial rates of rapid virologic response, although virological breakthroughs associated with resistance mutations were observed. This combination has been evaluated in multiple phase II studies, but is not approved or marketed.
