GSK 4-component Strep A vaccine (Alum-adjuvanted)

GSK 4-component Strep A vaccine (Alum-adjuvanted) is an investigational vaccine developed by GlaxoSmithKline (GSK) and the GSK Vaccines Institute for Global Health (GVGH) to prevent Group A Streptococcal (Strep A) infections. The vaccine is composed of four distinct antigens: three recombinant proteins—Streptolysin O (SLO), Streptococcus pyogenes Adhesion and Division protein (SpyAD), and Streptococcus pyogenes Cell Envelope Protease (SpyCEP)—and a glycoconjugate consisting of the Group A Carbohydrate (GAC) cell wall polysaccharide conjugated to a CRM197 carrier protein. This specific formulation is adjuvanted with aluminum hydroxide (Alum) to enhance the immune response. The vaccine aims to provide broad protection against various Strep A strains, thereby preventing superficial infections (such as pharyngitis and impetigo) and reducing the incidence of severe post-streptococcal complications, including acute rheumatic fever (ARF) and rheumatic heart disease (RHD). It is currently undergoing Phase 1 clinical evaluation for safety and immunogenicity.
