Overview

GSK2132231A + dacarbazine is a discontinued experimental **combination therapy** for cancer, specifically for unresectable/metastatic malignant melanoma. \n**GSK2132231A** (also known as astuprotimut-R, zastumotide, or MAGE-A3 ASCI) is a recombinant **MAGE-A3 antigen-specific cancer immunotherapeutic vaccine**, designed to stimulate an immune response against cells expressing the MAGE-A3 tumor antigen. It is formulated with the AS15 immunological adjuvant system to enhance immunogenicity. **Dacarbazine** is an established alkylating chemotherapy agent that causes DNA damage to cancer cells. \nThis combination was developed to target MAGE-A3-positive tumors (melanoma) and intended to harness both direct cytotoxic (dacarbazine) and tumor-targeted immunotherapeutic effects (GSK2132231A). The regimen was administered via intramuscular (GSK2132231A) and intravenous (dacarbazine) routes. \nThe development was discontinued after clinical trials failed to demonstrate sufficient clinical benefit.
