Overview

GSK2336805 is an investigational small molecule inhibitor targeting the hepatitis C virus (HCV) non-structural protein 5A (NS5A), a key component in HCV replication. Ritonavir is a protease inhibitor primarily used as a pharmacokinetic enhancer due to its potent inhibition of cytochrome P450 3A4 (CYP3A4), thereby increasing plasma concentrations of co-administered drugs metabolized by this enzyme. In combination, GSK2336805 and ritonavir are studied for their potential in treating HCV infection, with ritonavir boosting the exposure of GSK2336805 by inhibiting its metabolism. This combination has been evaluated in Phase I clinical trials for safety and pharmacokinetics in healthy subjects[2][3]. Ritonavir is also approved as an antiretroviral agent for HIV-1 infection[1][4][7].
