Overview

GSK2374697 is a genetically engineered fusion protein consisting of a human domain antibody (AlbudAb) fused to exendin-4, designed to bind with high affinity to human serum albumin. This albumin-binding property extends the systemic exposure and half-life of the drug, making it a long-duration glucagon-like peptide-1 (GLP-1) receptor agonist. The molecule was developed as an antihyperglycemic agent and obesity therapy, with pharmacological effects including reduction in postprandial glucose and insulin levels, delayed gastric emptying, and feedback suppression of endogenous GLP-1 and peptide YY secretion. Clinical studies demonstrated prolonged pharmacokinetics (half-life 6–10 days), expected GLP-1 agonist effects on glucose metabolism, and tolerability consistent with other potent GLP-1 agonists. The drug was investigated for potential use in type 2 diabetes mellitus and weight loss[1][2][5][6].
