GSK3174998 + pembrolizumab is an investigational immunotherapy combination that pairs GSK3174998, a humanized IgG1 anti-OX40 agonistic monoclonal antibody, with pembrolizumab (Keytruda), an anti-PD-1 therapy. The combination targets complementary immune pathways to enhance anti-tumor activity. GSK3174998 activates OX40 (CD134) on T-cells, promoting their proliferation and survival while blocking regulatory T-cell function. Pembrolizumab blocks PD-1 interaction with its ligands (PD-L1 and PD-L2), activating immune responses against cancer cells. This combination was developed by GSK and Merck to potentially improve outcomes for patients with advanced solid tumors.
