**GSK3745417 + dostarlimab** is an experimental combination of two investigational immuno-oncology agents, being studied for cancer. GSK3745417 is a synthetic small molecule *stimulator of interferon genes* (**STING**) agonist developed by GSK. It works by activating the STING pathway to induce production of type I interferons and pro-inflammatory cytokines, promoting innate and adaptive immune responses and resulting in direct tumor cell death and immune activation[2][3][4]. **Dostarlimab** is a monoclonal antibody and a programmed death-1 (**PD-1**) immune checkpoint inhibitor, also developed by GSK, that blocks PD-1 on T-cells, thereby preventing cancer cells from evading immune attack. The combination aims to synergistically enhance anti-tumor responses through both innate (via STING) and adaptive (via PD-1 blockade) immune activation.
