GSK4424989 + alum

GSK4424989 + alum is an investigational 4-component vaccine being developed by GSK to prevent Group A Streptococcus (Strep A) infections and their autoimmune sequelae, including Acute Rheumatic Fever (ARF) and Rheumatic Heart Disease (RHD). The vaccine's active components consist of three recombinant proteins—Streptolysin O (SLO), SpyAD, and SpyCEP—and one glycoconjugate comprising the Group A Carbohydrate (GAC) antigen conjugated to a recombinant CRM197 carrier protein. This specific formulation is adjuvanted with aluminum hydroxide (Alum) to enhance the immune response. It is currently in Phase 1 clinical development, where it is being evaluated for safety, reactogenicity, and immunogenicity in healthy adults. The vaccine is designed to elicit a protective immune response against key surface and secreted proteins of *Streptococcus pyogenes*.
