GSK5764227 + atezolizumab is an investigational combination therapy for cancer, particularly under evaluation for advanced solid tumors and extensive-stage small cell lung cancer (ES-SCLC). GSK5764227 is a B7-H3-targeted antibody-drug conjugate (ADC) developed by GSK that binds to the B7-H3 (CD276) antigen, delivering a cytotoxic payload selectively to B7-H3-expressing tumor cells, causing cell death. Atezolizumab is a monoclonal antibody and immune checkpoint inhibitor targeting programmed death-ligand 1 (PD-L1), reactivating T-cell anti-tumor activity by blocking PD-L1-mediated immunosuppression. The rationale for combining these agents is to enhance anti-tumor efficacy by both directly killing cancer cells with the ADC and boosting immune-mediated tumor destruction with checkpoint blockade. The combination is being studied in ongoing Phase 1/2 trials for relapsed/refractory osteosarcoma, extensive-stage small cell lung cancer, and other advanced solid tumors[1][2][3].
