Overview
GSK5764227 + cisplatin is a combination therapy consisting of **GSK5764227**, a B7-H3-targeted antibody-drug conjugate (ADC), and **cisplatin**, a platinum-based alkylating chemotherapy agent. GSK5764227 (formerly HS-20089) was licensed by GSK from Hansoh Pharma and features a humanized monoclonal antibody against B7-H3 (CD276) conjugated to a topoisomerase I inhibitor payload via a protease-cleavable linker. B7-H3 is an immune checkpoint member of the B7 family that is overexpressed in various solid tumors and associated with poor prognosis. Cisplatin induces DNA cross-linking, leading to strand breaks and apoptosis. The combination is being evaluated in the Phase 1 EMBOLD PanTumor-101 trial (NCT06551142) for participants with advanced solid tumors, including lung and gynecological cancers, to leverage potential synergistic effects between the targeted ADC and systemic chemotherapy.
