GT103 + pembrolizumab is a combination therapy being investigated for the treatment of refractory, metastatic non-small cell lung cancer (NSCLC). GT103 is a first-in-class, fully human, IgG3 monoclonal antibody that targets complement factor H (CFH), which is implicated in cancer immune evasion. CFH regulates C3 convertase in the alternative pathway of the complement cascade, limiting downstream opsonization and membrane attack complex formation. Inhibition of CFH by GT103 facilitates complement-dependent tumor cell lysis, modulates the adaptive immune response, and inhibits tumor growth. Pembrolizumab is an established anti-PD-1 immunotherapy. The combination is being studied to potentially enhance anti-tumor activity in patients who have progressed on prior therapies, including PD-1 treatment.
