Overview

This is a combination of two drugs used in the management of attention deficit hyperactivity disorder (ADHD). Guanfacine (SPD503, marketed as Intuniv) is a selective alpha-2A adrenergic receptor agonist and acts as a non-stimulant for ADHD. Its mechanism in ADHD is not fully understood but may involve enhancing executive function via action in the prefrontal cortex. Lisdexamfetamine (Vyvanse) is a central nervous system stimulant and prodrug of dextroamphetamine, which increases levels of norepinephrine and dopamine by promoting their release and blocking reuptake, thereby improving focus and impulse control. Both drugs are approved for use in children and adults with ADHD; lisdexamfetamine is also approved for moderate to severe binge eating disorder in adults[1][6][8]. Clinical studies have shown no clinically significant pharmacokinetic interaction when these drugs are co-administered[4].
