Overview

**guselkumab + golimumab** is an experimental combination therapy comprising two monoclonal antibodies, guselkumab and golimumab. Guselkumab selectively binds to the p19 subunit of interleukin-23 (IL-23), blocking its pro-inflammatory signaling, while golimumab is a tumor necrosis factor alpha (TNFα) inhibitor, neutralizing both soluble and transmembrane forms of TNFα. This combination is being investigated for the treatment of moderately-to-severely active ulcerative colitis. In clinical trials, combination therapy demonstrated higher efficacy than either monotherapy, with greater clinical, histologic, and endoscopic remission rates, and a safety profile comparable to monotherapies. Guselkumab is developed and manufactured by Janssen (a Johnson & Johnson company); golimumab is also developed by Janssen.
