This combination therapy regimen consists of the oncolytic virus H101 (Oncorine) and hepatic arterial infusion chemotherapy (HAIC) using the FOLFOX regimen (oxaliplatin, 5-fluorouracil, and leucovorin calcium). H101 is a recombinant human adenovirus type 5 with E1B-55K and partial E3 deletions, designed to selectively replicate in and lyse tumor cells with defective interferon-responsive antiviral pathways or p53 mutations. In this specific protocol for intrahepatic mass-forming cholangiocarcinoma, H101 is administered via intratumoral injection to prime the tumor environment, followed by regional chemotherapy delivered through the hepatic artery to maximize local cytotoxic effects while minimizing systemic toxicity. The study is being conducted by Beijing Tsinghua Chang Gung Hospital to evaluate the efficacy and safety of this combined approach in patients with unresectable disease.
