hAd5-S-Fusion+N-ETSD

**hAd5-S-Fusion+N-ETSD** is an investigational, second-generation human adenovirus serotype 5 COVID-19 vaccine developed by ImmunityBio. The E1-, E2b-, and E3-deleted hAd5 vector encodes a modified SARS-CoV-2 spike antigen designed to enhance spike surface expression and a nucleocapsid antigen fused to an Enhanced T-cell Stimulation Domain that directs nucleocapsid toward endosomal and lysosomal compartments to enhance antigen presentation. The vaccine is intended to induce spike-directed neutralizing antibodies plus spike- and nucleocapsid-specific T-cell responses, including cellular immunity against conserved nucleocapsid epitopes. It has been evaluated as a prophylactic primary vaccine and supplemental booster using subcutaneous, oral capsule, sublingual, and intranasal approaches; a registered Phase 2/3 COVID-19 study was conducted as the TCELL VACCINE TRIAL.
