Overview

HAIC + surufatinib + tislelizumab is a combination treatment regimen being investigated for the first-line treatment of unresectable or metastatic biliary tract cancer. The regimen consists of Hepatic Arterial Infusion Chemotherapy (HAIC) using the FOLFOX protocol (oxaliplatin, fluorouracil, and leucovorin), which delivers high concentrations of chemotherapy directly to the liver to maximize local control. This is combined with surufatinib, an oral small-molecule tyrosine kinase inhibitor that targets VEGFR1/2/3, FGFR1, and CSF-1R to inhibit angiogenesis and modulate the tumor microenvironment, and tislelizumab, a humanized monoclonal antibody that acts as an immune checkpoint inhibitor by targeting the PD-1 receptor. This triple-modality approach is designed to synergize local regional therapy with systemic anti-angiogenic and immune-mediated anti-tumor activity.
